Description
Mycoplasma hyopneumoniae real-time PCR test kit enables the safe detection of Mycoplasma hyopneumoniae DNA in respiratory tract secretion swab, lung tissue from pigs and bacterial cultures within 60 minutes. The kit consists of fluorescence PCR mix, positive control and negative control for real-time PCR amplification of Mycoplasma hyopneumoniae. Fluorescence PCR mix contains primers, probes, enzyme and buffer for detection of the Mycoplasma hyopneumoniae p36 gene.
[Sample Information]
Respiratory tract secretion swab,
Lung tissue and
Bacterial culturesserum(all need DNA extraction )
[Components]
Fluorescence PCR Mix | 1.2mL | - 2 0℃ |
Positive Control | 1 tube | - 2 0℃ |
Negative Control | 1 tube | - 2 0℃ |
Instruction manual | 1 box |
PCR STEP
(1) Take out the reagents in the room temperature for melting, mixing and vortex.
Centrifuge briefly.
(2) Dispense 20 µL of fluorescence PCR mix to the appropriate number of PCR strip
tubes.
(3) Add 5 μL of sample supernatant to each well (include the positive control and
negative control for each test). The final reaction volume should be 25 μL.
(4) Seal each PCR tube, then centrifuge briefly to bring the contents to the bottom of
the plate tubes.
(5) Write down the order of each sample.
(6) Set up PCR amplification cycling program as following:
Volume | 25 μL | |||
Signal channel | FAM signal channel to record fluorescence
Step | Temperatures | Time | Cycles
Amplification | 1 | 50℃ | 2min | 1 |
2 | 95℃ | 3min | 1 | |
3 | 96℃ | 10sec | 45 | |
56℃ | 20sec |
